The vagus nerve was injected with horseradish peroxidase
conjugated to wheat germ agglutinin (WGA-HRP) (Sigma), reconstituted
to 1% in distilled water, in a series of 15 ferrets.
Animals were fasted for 24 h prior to anesthesia. Ferrets were
preanesthetized with an intramuscular injection of a ketamine/
xylazine mixture (25 mgkg ketamine:2 mg/kg xylazine). They
then were intubated and maintained at a surgical plane of anesthesia
with halothane. The vagus nerve and nodose ganglion were
approached through a parasagittal incision. The carotid sheath
was visualized and opened, the vagus nerve identified, dissected
free from surrounding tissue, and elevated. The nerve and its
nodose ganglion were injected with 10 µl of a 1% WGA-HRP
solution in distilled water delivered through a 10 µl Hamilton
microsyringe through a puncture 2-3 cm distal to the nodose
ganglion. The needle of the microsyringe was threaded through
the nerve proximally to the level of the nodose ganglion where
five injections of 2 µl each were made within the substance of
the ganglion. |
